One of the major goals of immunotherapy is to achieve specific modulation of the immune system to combat malignancies without causing significant systemic reactions. One strategy for achieving this goal is the use of biodegradable polymers for controlled release of immune modulating agents such as cytokines or immune checkpoint inhibitors. We have recently shown that this approach can be used to effectively enhance immune responses against a range of different tumors without causing significant systemic reactions. Here we report on the development of a novel biodegradable polymer formulation for controlled release of IL-15 for use in immunotherapy. The polymer formulation was developed using a combination of solvent casting and solvent evaporation techniques and was characterized using dynamic light scattering (DLS), atomic force microscopy (AFM), scanning electron microscopy (SEM), high performance liquid chromatography (HPLC), fluorescence spectroscopy, differential scanning calorimetry (DSC) and water content measurements. The developed polymer formulation showed sustained release of IL-15 with initial burst release being observed followed by gradual release over several days in vitro. This study provides new insight into the potential use of biodegradable polymer formulations for controlled release of IL-15 for use in immunotherapy.The first time I saw this movie was when it was released on DVD and I was completely blown away by it. It was a movie that I had never heard of before and it was directed by a woman who had only directed one other movie before this one (the 2003 thriller "Down To Sleep").
